Deciphering complex immune pathways

Immune cells allow the human body to exist in a stable chemical and physical state (homeostasis) and have the ability to adapt as we develop and age. But changes to immune cell biology, for example, from chronic perturbations, alter homeostasis, leading to disease. To understand how and why these changes occur to our stable state, we must apply an interdisciplinary approach that combines physiology and immunology and need to decipher the complex immune pathways involved in cancer and chronic disease.

Seven Queen’s University researchers are contributing their knowledge in the areas of melanoma, intensive care unit survivors, postoperative pain, diabetes medication, Indigenous public health, and depression thanks to funding from the Canadian Institutes of Health Research.

Queen’s received a total of $3.95 million from the Fall 2019 CIHR Project Grant competition, a program that helps advance health-related research. With an eye on collaboration, the competition funds both individuals and groups of researchers at any career stage in all areas of health-related research.

Achieving Better Outcomes for Patients with Cancer

The results of clinical trials give us a sense of what is possible when it comes to new cancer treatments. But, there are many uncertainties around how these new treatments are being used, and how outcomes for patients compare with the results from clinical trials. Dr. Christopher Booth, Canada Research Chair in Population Cancer Care, wants to improve both cancer care and outcomes for Canadians by addressing these questions.

Dr. Janet Dancey is the Director of the Canadian Cancer Trials Group, also known as the CCTG, and a Professor in the Department of Oncology at Queen’s. She is also the Director of the High Impact Clinical Trials Program at the Ontario Institute for Cancer Research. The CCTG has over 80 member institutions, comprising over 2100 Canadian investigators who have facilitated over 500 trials in more than 40 countries. They are also the only non-American partner of the US National Clinical Trials Network.

My focus is cancer cell invasion and migration. This spheroid of cells is embedded in a 3D protein matrix and fluorescently stained for 3 proteins. Individual cells can be seen radiating away on all sides. The cells use integrin (red) and actin (green) to pull on the protein matrix and move, while cadherin (blue), binds and holds cells together within the spheroid. Cadherin normally holds clumps of cells together and allows them to form structures such as tissues and organs. This picture shows how invading cells lose their cadherin and express large amounts of integrin and actin, and it demonstrates some of the protein expression changes that cancer cells undergo which allow them to invade away from a tumour.
