Innate Immune Memory in Asthma and the Quest for Targeted Immunotherapies

Dr. Eva Kaufmann’s research explores a new concept in innate immunity, termed "innate immune memory", or "trained immunity". Her lab studies whether allergen exposure induces long-lasting epigenetic changes in hematopoietic stem cells and innate immune cells, such as monocytes. Such epigenetic reprogramming can be a reason why asthmatic individuals experience exacerbations and are more susceptible to severe clinical trajectories of respiratory infections.

Deciphering complex immune pathways

Immune cells allow the human body to exist in a stable chemical and physical state (homeostasis) and have the ability to adapt as we develop and age. But changes to immune cell biology, for example, from chronic perturbations, alter homeostasis, leading to disease. To understand how and why these changes occur to our stable state, we must apply an interdisciplinary approach that combines physiology and immunology and need to decipher the complex immune pathways involved in cancer and chronic disease.

The cornea is one of the most densely innervated tissues in the human body. The high presence of sensory nerve endings makes the cornea extremely sensitive to pain and changes at the eye’s surface. This is particularly important in regulating tear production, a function which is impaired in dry eye disease. By analyzing changes in nerve patterns and ion channel expression, we aim to further clarify the role of corneal nerves in spontaneous pain and tear production in dry eye disease.
